A collaboration between the Ando Lab, Penn State University and MIT takes a look at how ribonucleotide reductases (RNRs) – the enzymes that make the building blocks needed for DNA biosynthesis – are regulated. Using small-angle X-ray scattering (SAXS), they propose a model to rationalize how structural changes in the enzyme's domains lead to its inhibition. The Mo (Seyedsayamdost) Lab presents the total chemoenzymatic synthesis of the aglycone version of vancomycin, the so-called last-resort antibiotic to treat bacterial infections that resist other drugs. By chemically synthesizing the precursor backbone, they then utilize enzymes of the Oxy family to form the bis-aryl ether linkages between rings C-D and D-E (blue and red) and the unprecedented enzymatically-installed biaryl crosslink between rings A-B (green). Using this platform, they also synthesize an unnatural derivative of vancomycin that is effective against vancomycin-resistant pathogens. A new collaboration between the Groves Lab and the molecular imaging departments at Merck and Mass General Hospital brings a novel method to replace unactivated aliphatic C-H bonds with fluorine-18 (18F), an isotope of fluorine commonly used for PET imaging. Using a manganese porphyrin catalyst, they are able to fluorinate a broad collection of molecules, including several bioactive compounds using a fluoride (18F-) instead of the electrophilic sources commonly used. The lab of Prof. David MacMillan at the Merck Center for Catalysis at Princeton presents yet another accomplishment of photoredox and nickel catalysis, this time on the merger of open-shell and closed-shell reactive pathways to functionalize alkynes. By leveraging the readiness of nickel to participate in 1- and 2-electron chemistries, they can engage an alkyl radical (generated by an open-shell pathway) and an alkyne (more suitable for closed-shell mechanisms) to afford geometrically defined olefins. Chemical modifications in the DNA and histones play a central role in regulating the extent of gene expression.
